No keywords in content

Given the fact that Google always coax on having unique and relevant on-page content, so can a web page rank for a keyword having no mention in the content especially when it`s competing with tier 1 companies. Will building back-links and content marketing be enough? Also by doing so will it fall under negative SEO?
